Niflumic acid is a nonsteroidal anti-inflammatory drug (NSAID) that inhibits the function of cyclooxygenase and 5-lipoxygenase. It has been shown to inhibit P-glycoprotein (P-gp) in vitro. Inhibition of P-gp leads to an increase in the intracellular levels of Ca2+ and activation of the ryanodine receptor, which is a protein found on the endoplasmic reticulum. This results in an increase in cellular excitability, which can lead to cell death. Niflumic acid also inhibits picolinic acid and pyrazinoic acid production, which are toxic metabolites that may play a role in its anti-inflammatory effects.
