Polyphyllin B is a natural compound that is the main constituent of Formosanin C, which has been used in traditional Chinese medicine for centuries. Polyphyllin B inhibits the activity of DNA polymerase, thus inhibiting the transcription and replication of DNA. It also binds to the bcl-2 protein and inhibits apoptosis by blocking the activation of caspases. This drug also blocks tumor necrosis factor-α (TNF-α) from binding to its receptor on cells. The anti-cancer effects are due to its ability to inhibit cyclin D2 expression, which prevents the progression through G1 phase and into S phase. In addition, it has been shown to be effective against oxidative injury in a rat model of acute toxicity.
