Propidium iodide is an intercalating dye that non-specially binds to double stranded DNA. Unbound dye has an excitation and emission wavelength maxima of 493nm and 636nm, respectively. Once bound to DNA, there is a shift in the excitation and emission wavelength maxima to 535nm and 617nm, respectively, causing an increase in fluorescence signal (20â30-fold). It is not permeable to intact cell membranes. Therefore, it is often used to stain dead cells, where it can penetrate the damaged cell membrane and stain cells red. In combination with SYTO 9, it can be used to estimate dead and viable cells in one staining step. It can also be used to quantify DNA in flow cytometry analysis and as a reporter in LAMP diagnostics protocols.
