Phenyl 2-chloroethyl ether is a chlorinated derivative of the chemical compound endoxifen. It is a potent antiestrogen, with IC50 values in the nanomolar range. Phenyl 2-chloroethyl ether has been shown to inhibit cell proliferation in vitro and in vivo by blocking estrogen receptors and inducing apoptotic cell death. The drug also has an inorganic acid terminus, which prevents it from being metabolized by esterases or glucuronidases, leading to its prolonged therapeutic effect.
Phenyl 2-chloroethyl ether is used clinically as a cancer chemotherapeutic agent. It targets cancer cells that depend on estrogen for growth by binding to the estrogen receptor alpha (ERα) and blocking estrogen signaling pathways, which leads to tumor shrinkage. This drug also inhibits the uptake of cancer drugs such as doxorubicin into cells and prevents the synthesis of DNA and RNA.
