Phorate is a chemical that inhibits the mitochondrial membrane potential and disrupts the production of ATP, resulting in cell death. The inhibition of ATP production by phorate is caused by its binding to the enzyme complexes in the electron transport chain. Phorate has been shown to cause liver lesions at high doses, and causes toxicity at lower doses. Phorate binds to sulfhydryl groups in proteins, forming a disulfide bond, which may be responsible for its toxicity. The polymerase chain reaction (PCR) technique has been used to detect phorate-resistant strains of bacteria from human serum samples. These resistant strains have been found to carry mutations in genes encoding enzymes that are targeted by phorate.
