    Description:1,10-Phenanthroline-5,6-dione (PD) is a molecule that binds to the active site of many enzymes and inhibits their activity. PD has been shown to have antimicrobial activity against a number of bacteria and fungi. In vitro assays have shown that PD is effective against tumor cells in carcinoma cell lines. In vivo studies have also shown that PD can inhibit the growth of tumors in mice by inhibiting DNA synthesis. It has been found that PD competes with the cofactor NADH for binding to the enzyme dihydrofolate reductase, which is required for DNA synthesis. This inhibition leads to an increased amount of hydrogen peroxide, which reacts with iron ions in the enzyme ferritin, converting it into ferric hydroxide. The redox potentials of this reaction are reversed when NAD+ is added to the reaction mixture, leading to regeneration of NAD+ and allowing DNA synthesis to resume.
