Sinomenine is a plant alkaloid that has been used in traditional Chinese medicine for centuries. It has anti-inflammatory and antiallergic activities, and may also have some analgesic properties. Sinomenine inhibits the production of inflammatory cytokines such as interleukin-1 (IL-1), IL-6, IL-8, and tumor necrosis factor (TNF)-α. Sinomenine is able to inhibit NF-κB binding activity by competitively inhibiting the binding of p65 to its DNA target sequence. Sinomenine also binds to the α7 nicotinic acetylcholine receptor and blocks signal pathways involved in cell proliferation, differentiation, and apoptosis. The optimum concentration of sinomenine hydrochloride for inhibition of NF-κB binding activity was found to be 0.5 μM with a constant pressure of 1 atm.
