Sulindac sulfone is a nonsteroidal anti-inflammatory drug that belongs to the class of sulfones. It has been shown to inhibit the mcl-1 protein, which is involved in the mitochondrial apoptosis pathway. Sulindac sulfone inhibits prostaglandin E2 (PGE2) levels and mitochondrial membrane potential in vitro assays. Sulindac sulfone also has inhibitory properties against a number of enzymes including cyclooxygenase-2 (COX-2), lipoxygenases, phospholipases A2, and thromboxane synthase. These properties are responsible for its anti-inflammatory effects. Sulindac sulfone shows minimal toxicity due to its low bioavailability and lack of liver metabolism. This drug has been shown to have anticancer activity as well as an ability to inhibit infectious diseases such as Leishmania major and Helicobacter pylori.
