Tenofovir is an antiviral drug that belongs to the class of acyclic nucleoside phosphonates. Tenofovir is a prodrug that needs to be activated by phosphorylation before it can act as a polymerase chain inhibitor. The inhibition of viral replication by tenofovir has been shown in many different models, including in murine sarcoma virus, where the drug was able to inhibit the growth of the virus in cell cultures and mice. Tenofovir also inhibits human HL-60 cells and has been shown to have anti-inflammatory properties, which may be due to its ability to inhibit prostaglandin synthesis. Structural analysis has revealed that tenofovir binds to HIV protease and prevents it from cleaving the polyprotein precursor into individual proteins. This leads to a decrease in HIV levels in both CD4+ and CD8+ T cells.
