Description:Timolol is an ophthalmic drug used for the treatment of glaucoma and other eye conditions, such as iritis. It belongs to the group of beta-adrenergic receptor antagonists and has a long history of use in humans and animals. Impurities in timolol include D-Timolol maleate, which is a metabolite that is formed by oxidation. Timolol impurity D (TMD) has been found to be more potent than timolol for inhibiting anhydrase activity in vitro. TMD also has a higher affinity for benzalkonium chloride than timolol, which may be due to its ability to form hydrogen bonds with this preservative. TMD profiles have been generated using chromatography and mass spectrometry techniques and show systematic changes in the concentration of TMD with increasing pH values. This profile shows that the thermodynamic stability of the compound increases as pH rises but that it does not change appreciably at neutral
