2,4,6-Trichloroanisole (TCA) is a molecule that is present in the environment and can be found as a contaminant in food and beverages, especially alcoholic beverages. TCA has been shown to act as an allosteric activator of the nicotinic acetylcholine receptor (nAChR) in mammalian cells. The binding of TCA to nAChRs leads to a change in the shape of the receptor, which changes the local concentration of ions around the cell membrane. This change causes an increase in permeability and leads to a decrease in intracellular potassium concentrations. TCA has also been shown to affect bacterial growth by inhibiting DNA synthesis through its effect on DNA gyrase or topoisomerase IV.
