This binding blocks the initiation reaction and prevents translocation of tRNA from the A site to the P site. The inhibition constant for 2,4,4-Trimethyl-2-pentanol was determined to be 0.012 mM using gel chromatography with acetate extract from human urine and rat kidney homogenates.
