Tafenoquine succinate is a drug that inhibits the polymerase chain reaction, which is an enzyme activity that replicates DNA. It is also used to treat malaria and leishmaniasis. Tafenoquine succinate belongs to the class of drugs called quinoline antimalarials. It has been shown to be safe in humans, with no serious side effects. Tafenoquine succinate binds to the mitochondrial membrane potential, which prevents the accumulation of proton gradients across the inner mitochondrial membrane. This leads to an increase in intracellular calcium levels, which can cause cell death due to oxidative stress. Tafenoquine succinate also inhibits nuclear DNA replication and bacterial growth by preventing the transcription of rRNA, leading to inhibition of protein synthesis and cell division.
