TFFH is a fluoroformamidinium salt used as a coupling reagent for amide formation and peptide synthesis. TFFH is a non-hygroscopic and stable salt, readily available, affordable and efficient. TFFH allows high yields in liquid and solid phase, in presence of sterically hindered carboxylic acids and hindered or electron-deficient amines, with minimum racemisation. The carboxylic acid is activated by TFFH forming the more reactive acyl fluoride, which is reportedly less prone to racemisation than the corresponding acyl chloride.
