o-Tolidine is an organic compound that is used in analytical chemistry as a reagent for the detection of amines. It reacts with amines to form an intensely coloured compound, which can be detected by a fluorescence detector. o-Tolidine has been shown to have a high toxicity in rats and humans, with no known antidote or treatment. The mechanism of this reaction is not yet fully understood, but it is believed to involve an acid-base reaction between the o-tolidine molecule and the amine at pH 2.5, followed by protonation of the o-tolidine molecule.
