2,4,4-Trimethylcyclopentanone is a cycloalkanone that has been identified as an off-flavor in wines. It is formed by the oxidation of 2,4-dimethylcyclohexane-1-carboxylic acid. The biosynthetic pathway of 2,4,4-trimethylcyclopentanone begins with the conversion of L-phenylalanine to cinnamic acid by phenylalanine ammonia lyase. Cinnamic acid is then converted to 4-coumaric acid by 4-coumarate CoA ligase. 4-Coumaric acid can be converted to ferulic acid by feruloyl CoA reductase and then to 2,4,4-trimethylcyclopentanone by oxidoreductases such as cytochrome P450 monooxygenases and alcohol dehydrogenases.
