Valencene is a natural compound that belongs to the class of sesquiterpene lactones. It can be found in Valerian, an herbaceous plant used medicinally in Europe since ancient times. Valencene has been shown to have antifungal activity in vitro and significant cytotoxicity. This compound is metabolized through a number of metabolic transformations, including oxidation by cytochrome P450 enzymes, reduction by glutathione reductase, or conjugation with glucuronic acid. Valencene also specifically binds to markers expressed at high levels in Mycobacterium tuberculosis strains (e.g., ESX-1 secretion system protein) and inhibits cell growth in culture. The reaction mechanism for valencene is not yet well understood; however, it has been proposed that it may involve oxygenated intermediates generated from the reaction of valencene with the enzyme polymerase chain reaction (PCR).
