6-Chloropurine is a purine derivative that is used as a precursor to the nucleotide base, thymine. It has been shown to be mutagenic in bacteria and in mammalian cells. 6-Chloropurine can be metabolized by the enzyme orotate phosphoribosyltransferase (OPRT) and converted into orotic acid. This reaction is irreversible, so it is important not to administer this drug in cases of hyperuricemia or gout. 6-Chloropurine also has labile properties and is easily degraded when exposed to light or air. The tendency for this drug to form tautomeric pairs with other bases, such as guanine or uracil, is what makes it mutagenic.
