AZD3463 is a small molecule that inhibits the ubiquitin proteasome. It has been shown to be effective in vivo, as well as in vitro, for the treatment of cancer. AZD3463 binds to the carbonyl group on the protein substrate and inhibits the activity of the drug-metabolizing enzyme, adp-ribose hydrolase. This inhibition prevents degradative processes from occurring within cells and leads to an increase in autophagy. AZD3463 also inhibits epidermal growth factor receptor (EGFR) signaling pathways and activates tumor suppressor genes such as p53 and p21. Pharmacokinetic properties have been studied in mice, rats, and cynomolgus monkeys.
