4'-Ethynyl-2'-deoxyadenosine (4EDA) inhibits viral polymerase and is a potent inhibitor of the influenza virus. 4EDA is an antiviral drug that has been shown to inhibit influenza virus replication in cell culture by inhibiting viral RNA synthesis. 4EDA blocks the enzymatic activity of influenza virus polymerase, which leads to inhibition of viral DNA synthesis and subsequent inhibition of protein synthesis. Resistance to 4EDA has been found to be associated with mutations in the linker region between the nucleotide binding site and the active site of the polymerase enzyme. This mutation prevents 4EDA from binding to the critical catalytic residue in this region, preventing its inhibitory effect on viral RNA synthesis.
