GNE-0723 is a novel allosteric modulator that enhances synaptic function. GNE-0723 binds to the NMDA receptor and potentiates the activity of glutamate, an excitatory neurotransmitter that plays a major role in synaptic plasticity. This drug has been shown to produce antidepressant-like effects in mice, which may be due to its ability to increase levels of brain-derived neurotrophic factor (BDNF) and synaptogenesis.
