TN-16 is a synthetic taxane with a high affinity for the microtubule polymerization protein, taxol. It binds to the β-taxol and prevents it from binding to the ɣ-taxol, which disrupts the formation of the polymerized microtubules. TN-16 is also reversible, meaning that it can be displaced by nocodazole or podophyllotoxin. In addition, TN-16 has been shown to inhibit cell proliferation in ovary cells and metaphase chromosomes in cell culture experiments. This drug also has shown anti-tumor activity in mice when used in combination with vinblastine or daunorubicin. TN-16 has cross resistance with other drugs such as vinca alkaloids, which are plant alkaloids that have been used to treat various cancers since they were first isolated from plants in Madagascar in 1939.
