Olvanil is a synthetic vanilloid compound that functions as an agonist at vanilloid receptors. It has been shown to inhibit the release of bowel movement in response to capsaicin, and has been used clinically for the treatment of bowel disease. Olvanil also inhibits nociception by acting on TRPV1 channels, and is being investigated as a potential treatment for chronic pain. Olvanil binds to capsaicin-sensitive neurons and causes a depolarization in these cells, which leads to the activation of voltage-gated calcium channels. This increases cytosolic calcium levels, which may lead to inhibition of neurotransmitter release or even neuronal death. Olvanil also binds to vanilloid receptors found on sensory neurons and blocks their activation by capsaicin or heat, preventing the sensation of pain.
