GNE-493 is a synthetic compound that is currently under development for its potential as a cancer biomarker and therapeutic agent. GNE-493 binds to the pro-apoptotic protein Bax, which is located in the mitochondrial membrane. This binding induces Bax oligomerization and release of cytochrome C from the mitochondria into the cytosol, where it activates caspases and triggers apoptosis. GNE-493 has been shown to induce cell death in several types of cancer cells (e.g., miapaca-2 cells) and potent antitumor activity in mice bearing human colon carcinoma xenografts. GNE-493 also inhibits tumor growth by inhibiting cyclin D1 expression and decreasing the mitochondrial membrane potential, leading to cell cycle arrest at the G0/G1 checkpoint.
