It has been shown to inhibit the hepatitis C virus (HCV) NS3 protease by binding to the active site of the enzyme. Boceprevir metabolite M4-d9 methyl ester binds to the HCV NS3 protease with a Kd of 1.2 nM and an IC50 of 2.6 μM in vitro. This peptide also inhibits HIV-1 entry into host cells by competitively inhibiting gp41 binding to CD4 or CCR5 receptors, thereby preventing viral fusion with host cell membranes.
