Teriflunomide is a drug that has been clinically proven for the treatment of multiple sclerosis. It is a prodrug that is converted to its active form, teriflunomide-phosphate, in vivo. Teriflunomide inhibits the activity of matrix metalloproteinases by blocking Ca2+ channels and inhibiting Ca2+ influx into the cytosol. This blocks the degradation of extracellular matrix proteins and reduces inflammation, which may lead to long-term efficacy in autoimmune diseases such as multiple sclerosis. Teriflunomide also decreases T-cell proliferation by inhibiting integrin receptors on T cells and monoclonal antibody production.
