Poly(ADP-ribose) glycohydrolase (PARG) is an enzyme that catalyzes the hydrolysis of poly(ADP-ribose). PARG is a key mediator of caspase-independent cell death. PARG inhibition has been shown to induce synergistic cell death in combination with other anti-cancer agents, such as histone deacetylase inhibitors and proteasome inhibitors. The drug can be used as a chemotherapeutic agent for bowel disease and experimental models. Poly(ADP-ribose) glycohydrolase has been shown to play a role in transcriptional regulation, x-ray crystal structures, and energy metabolism. When PARG is inhibited, cytosolic calcium levels increase, which can lead to the activation of calpain and subsequent cleavage of cytoskeletal proteins.
