SKI 178 is a potent and selective inhibitor of the enzyme soluble epidermal growth factor receptor, which is involved in cell signaling. This drug has been shown to inhibit the proliferation of tumor cells in solid tumours, lymphoproliferative disorders, and leukemias. SKI 178 inhibits the activation of epidermal growth factor receptor-mediated downstream signaling pathways that are required for cellular proliferation and survival. In addition, it induces apoptosis by activating caspase-3 in cancer cells. The molecular mechanisms of SKI 178's anti-tumor activity have been elucidated through genetic ablation experiments as well as biochemical properties. SKI 178 is a small molecule that binds to the heparan sulfate proteoglycan on the surface of cells, thereby inhibiting epidermal growth factor from binding to its receptor on the cell surface.
