N-Fmoc-D-alaninehydrate is a synthetic, chiral, and asymmetric compound that has been synthesized to mimic the natural product N-acetylhistone deacetylase inhibitor. It is a macrocyclic molecule composed of two quinazolones connected with an ansamycin core. The stereogenic center in the ansamycin core has been shown to be crucial for activity. It has been found that N-Fmoc-D-alaninehydrate binds to histone deacetylases (HDACs) and inhibits their enzymatic activity by binding to their active site. This inhibition leads to decreased levels of acetylation of histones, which is associated with increased transcription of genes that are important for cell growth and proliferation.
