PX-478 is a drug that inhibits the function of P-glycoprotein (P-gp), which is a protein that transports certain drugs out of cells. PX-478 has been shown to inhibit the proliferation of tumor cells in vitro in response to radiation. It also inhibits the HIF-1α pathway, which leads to apoptosis and prevents tumor growth. In vivo, PX-478 inhibited tumor growth and prolonged survival time in a squamous carcinoma model system. The drug binds to the DNA response element upstream of the polymerase chain gene, thereby inhibiting transcription and translation.
