KY-556 is a peptide inhibitor of the Kv1.2 voltage-gated potassium channel. It is a potent and selective blocker of Kv1.2 channels, with an IC50 of 0.4 μM and no activity against other voltage-gated potassium channels (Kv1.1, 1.3, 1.6). KY-556 has been shown to inhibit the proliferation of tumor cells by blocking Kv1.2 channels in cell culture experiments and in vivo in mice with implanted melanoma tumors. KY-556 also inhibits the release of inflammatory cytokines such as IL-6 and TNFα from human monocytes and macrophages, suggesting that it may have anti-inflammatory properties as well as tumor suppression capabilities.
KY-556 is synthesized using solid phase peptide synthesis on a Rink amide resin with an N-terminal lysine side chain protecting group and a COOH terminus acid side chain protecting group.
