Pevonedistat is a sulfamate, dihydro, nedd8-activating enzyme inhibitor that has been shown to inhibit the activity of NEDD8. Pevonedistat is used in cancer chemotherapy and prevents methylation of DNA by inhibiting the activity of enzymes that activate NEDD8. Pevonedistat has also been shown to be a marker for mesenchymal stem cells and can be used to differentiate between these cells and other types of cells. The drug binds to sulfamic acid, an enzyme inhibitor, and blocks the conversion of hypomethylated cytosine into methylcytosine. This causes hypomethylation, which leads to DNA damage. Hypomethylation also leads to mutations in cancer cells, making them more susceptible to death by chemotherapeutic agents such as 5-fluorouracil or cisplatin.
