AMG 1694 is a cytosolic inhibitor of the glucose-6-phosphatase enzyme, which has been shown to act as an antidiabetic drug in animal models. AMG 1694 has been shown to have a potent inhibitory effect on gluconeogenic enzymes in vitro and in vivo. In addition, AMG 1694 has a high uptake by cells, with minimal systemic pharmacokinetic effects. This molecule also inhibits the regulatory protein phosphofructokinase (PFK), which is involved in the metabolic pathway for glycolysis. AMG 1694 also inhibits the enzyme fructose-1,6-bisphosphatase, which regulates glucose metabolism. This agent is currently being studied for its use as an antidiabetic drug in humans.
