Nav1.7 is a voltage-gated ion channel that is important for the transmission of pain signals. Nav1.7 blockers are designed to inhibit Nav1.7 with an IC50 value of ~0.4 μM, and can be used in the treatment of chronic pain associated with inflammatory conditions, such as arthritis and fibromyalgia, or cancer-related pain. These drugs have been shown to inhibit the activity of Nav1.7 by binding to residues in its activation gate, preventing the opening of sodium channels at resting membrane potentials and blocking the propagation of action potentials in peripheral sensory neurons, thereby inhibiting nociceptive neurotransmission.
