2,2'-Anhydrothymidine is a synthetic nucleoside that has been shown to inhibit the synthesis of DNA and RNA in vitro. It is also an inhibitor of acyclovir-resistant herpes simplex virus and can be used to treat infections caused by this virus. 2,2'-Anhydrothymidine can be synthesized from ethylene glycol and formaldehyde. This compound has affinity for purine and pyrimidine nucleosides, which are present in DNA and RNA. 2,2'-Anhydrothymidine has a conformation that allows it to bind to the nucleoside at the alpha-carbon atom in the ribose ring. This conformation is stabilized by hydrogen bonding with the oxygen atoms on adjacent carbons in the same ring or adjacent rings. The presence of acid catalysts increases yields of this compound, although yields with alkaline catalysts are increased at higher temperatures.
