2',3',5'-Tri-O-acetyluridine (TAU) is a triacetate prodrug of uridine that is used as an antidote for diabetic neuropathy and inflammatory bowel disease. TAU has also been shown to be effective in the treatment of metastatic colorectal cancer. The drug interactions with TAU include erythromycin, which inhibits its metabolism, and cimetidine, which inhibits its absorption. TAU is metabolized by orotic aciduria to orotic acid and then excreted in the urine. The symptoms of TAU poisoning are nausea, vomiting, diarrhea, fatigue, fever, hypotension, and convulsions. TAU can cause orotic aciduria in children when given intravenously at high doses. The histological analysis of TAU shows that it does not accumulate in tissues or cells because it is rapidly hydrolyzed to uridine by cytosolic enzymes before leaving the cell.
