4'-Thioadenosine is a nucleoside that has been shown to inhibit the growth of leukemia cells by binding to adenosine receptors. It is also able to inhibit the activity of the mammalian phosphodiesterase PDE4, which is responsible for hydrolyzing cAMP. The cellular effects of 4'-thioadenosine can be seen in HL-60 human cells, and this drug has also shown promising results in animal models of kidney fibrosis and liver cirrhosis. 4'-Thioadenosine has been shown to have anti-inflammatory properties, which may be due to its ability to inhibit prostaglandin synthesis.
4'-Thioadenosine's inhibition of adenosine receptor activity may lead to negative side-effects such as severe bronchoconstriction or cardiac arrhythmias.
