Nucleoside phosphoramidites are used in the chemical synthesis of oligonucleotides. N6-Benzoyl-2'-deoxy-5'-O-DMT-2'-fluoroadenosine 3'-CE phosphoramidite, also known as 2'-fluoro-dA CEP, is a modified adenosine with a fluorine atom in the 2’ position to enable the synthesis of non-natural fluorine containing oligonucleotides. Nucleoside phosphoramidites are typically modified by displacement of the diisopropylamine group with the 5’ hydroxyl of the next nucleoside in the presence of an azole catalyst such as 1H-tetrazole or 4,5-dicyanoimidazole. 2'-Fluoro-dA CEP has been used in the synthesis of phosphorothioate oligonucleotides that were highly nuclease resistant yet retained exceptional binding affinity to the RNA targets.
