Description:PF-06297470 is a novel, potent and selective allosteric modulator of the metabotropic glutamate receptor type 1 (mGluR1). It has been shown to be orally active in animal models. This drug has been shown to have anti-Parkinsonian effects, which are related to its ability to inhibit glutamate release. PF-06297470 also inhibits the binding of glutamate to mGluR1 receptors, thereby inhibiting the activation of these receptors and reducing the transmission of excitatory signals in the brain. PF-06297470 binds with high affinity (Ki=0.5 nM) and selectivity to mGluR1 receptors, with no appreciable affinity for other metabotropic glutamate receptors or ion channels at concentrations up to 10 μM.
