1,6,6-trimethyl-6,7,8,9-tetrahydrophenanthro[1,2-b]furan-10,11-dione (TMTD) is a cardiotoxic drug that has been shown to be an inducer of cardiac apoptosis. TMTD is a proapoptotic agent that induces the death of neurons in vivo and in vitro. The effects of TMTD on neurons are mediated by activation of caspase 3 and caspase 9. TMTD also activates GSH peroxidase activities and reduces the extent of atherosclerotic lesions. It also inhibits glucose uptake into cells and can lower blood sugar levels. These effects may be due to its ability to inhibit insulin release from pancreatic β cells or its ability to inhibit glucose transport into cells.
