Rosoxacin hemisulfate (ROX) is a fluoroquinolone antibiotic that inhibits bacterial DNA gyrase and topoisomerase IV. ROX has been shown to be active against gram-positive bacteria, including Enterococcus faecium and Streptococcus pyogenes. It also has activity against gram-negative bacteria, such as Escherichia coli K-12 isolates and Proteus mirabilis isolates. ROX is selectively toxic for gonorrheal organisms by inhibiting the growth of resistant mutants. Rosoxacin hemisulfate has been shown to be effective in treating choroidal neovascularization in an experimental model. The mechanism of action of ROX is similar to other quinolones: it inhibits bacterial DNA synthesis by binding to the enzyme DNA gyrase which leads to inhibition of bacterial growth.
