4-Hydroxy-1-(methylsulphonyl)piperidine is a novel linker molecule that has been designed to be orally bioavailable. It is hydrophobic and has been optimized for interaction with protein-coupled receptors. This linker molecule has shown excellent plasma glucose lowering effects in animal studies, which may be due to its ability to reduce insulin resistance and increase insulin sensitivity. The conformational and conformationally dependent interactions of this compound with the receptor are expected to improve oral bioavailability. 4-Hydroxy-1-(methylsulphonyl)piperidine also inhibits the activity of protein kinase C (PKC), although it does not inhibit phosphodiesterase or adenylate cyclase activities.
