Tetrazine-NHS ester (TNT) is a positron-emitting probe that has been used to study the distribution of tumor cells. It is taken up by cells in proportion to their affinity for the TNT receptor and can be detected with a PET scan. Immunoconjugates of TNT have been developed for use in tumor therapy. The TNT-antibody conjugate can be directed against specific tumor cells, such as colorectal carcinoma, and then activated by radiation or cytotoxic drugs. The TNT-immunoconjugate is taken up by the targeted tumor cell and subsequently destroys it through an intracellular process involving DNA ligation and transduction.
