GSK1016790A is a potent and selective small molecule inhibitor of TRPV4, a member of the transient receptor potential (TRP) channel family. It is currently being investigated for pharmacological effects in vitro and in vivo. GSK1016790A has been shown to inhibit the activity of TRPV4 channels in neuronal cells, leading to an increase in intracellular calcium levels and subsequent neuronal death. This agent also blocks the angiotensin system, which may have physiological implications including regulation of blood pressure. GSK1016790A binds to calcium binding sites on the cytosolic side of TRPV4 channels where it acts as a competitive antagonist. The compound is stable over a wide pH range and does not show any significant effect on cellular viability at concentrations up to 100 µM.
