Compound 401 is a synthetic inhibitor of the protein kinase s6 (PKS6) that has been shown to have anti-cancer activity. It was found to inhibit cancer stem cells in vitro and tumor growth in vivo. The mechanism of action for Compound 401 is not yet known, but it is thought to be a pathway inhibitor because it blocks the activation of PKS6 by binding to its active site. The tautomeric form of Compound 401 is an aspartyl molecule that binds to the active site of PKS6, preventing its phosphorylation by protein kinase A. This inhibition prevents the phosphorylation of serine residue 797, which is required for transcriptional elongation. Compound 401 has also been shown to have antiviral properties against hepatitis B virus and human immunodeficiency virus type 1 (HIV-1).
