NVP-TNKS656 is a small molecule that inhibits β-catenin in the microenvironment of colorectal cancer. β-catenin is an enzyme that helps regulate the Wnt pathway, which plays a role in tumor progression. NVP-TNKS656 binds to the tyrosine kinase domain of β-catenin and inhibits its activity by blocking the binding site for ATP. It also binds to other proteins involved in Wnt signaling, such as ARMET and GSK3β. This inhibition disrupts the normal function of these enzymes and prevents them from phosphorylating downstream targets, resulting in reduced cell proliferation and migration. NVP-TNKS656 has been shown to inhibit mouse tumors in vivo and reduces bone cancer incidence in mice with a genetic predisposition for this type of cancer.
