Celecoxib-d7 is a drug that is metabolized by the cytochrome P450 system. It is an atypical compound that produces a concentration-dependent inhibition of human liver cytochrome P450. Celecoxib-d7 has been shown to inhibit the metabolism of other drugs, such as indomethacin, phenacetin, and sulfamethoxazole, which are substrates for CYP1A2. The matrix effect can be controlled by the addition of a small amount of quinidine to the incubation medium. This drug also binds allosterically to the enzyme cytochrome P450, which leads to changes in its kinetics and molecular modeling properties. The clinical studies have shown that celecoxib-d7 affects plasma concentrations in a dose-proportional manner and does not have any significant effects on plasma protein binding.
