BMS-345541 is a signal transduction inhibitor that blocks the IKK enzyme, which is involved in inflammatory signaling. Studies have shown that BMS-345541 has an anti-inflammatory effect and significant cytotoxicity against cancer cells. BMS-345541 also inhibits the activity of Toll-like receptor 2 (TLR2) and TLR4, which are proteins on the surface of cells that recognize microbes. These receptors initiate signaling pathways that activate NF-κB, a transcription factor necessary for inflammatory response. BMS-345541 binds to DNA and prevents it from being transcribed into RNA, inhibiting cellular transformation. The drug also inhibits polymerase chain reactions by binding to the catalytic subunit of DNA polymerase. It has been shown to inhibit cell proliferation in vitro as well as in vivo by blocking Ca2+ uptake into cytoplasmatic organelles. Furthermore, it has been shown to be effective for treating bowel disease by reducing
