FICZ is a regulatory protein that belongs to the family of transcription factors. FICZ is synthesized in cells as a precursor peptide and is cleaved into two subunits, an N-terminal alpha-helical domain and a C-terminal beta-sheet domain. The N-terminal alpha-helical domain consists of six helices and interacts with DNA by binding to the major groove of the double helix. The C-terminal beta-sheet domain interacts with RNA polymerase II, which is responsible for transcription of DNA into RNA. FICZ has been shown to inhibit inflammatory bowel disease, skin cancer, and autoimmune diseases. This protein also has been shown to inhibit inflammation in animal models through growth factor activity and oxidative DNA damage.
