Isoflavan is a natural compound found in the acetate extract of the roots of Isoflavus. It has biological properties that inhibit autoimmune diseases and has been shown to have antioxidant, anti-inflammatory, and immunosuppressive activities. Isoflavan binds to DNA in a noncompetitive manner and inhibits specific mitochondrial enzymes involved in mitochondrial function. This binding prevents the formation of reactive oxygen species (ROS) by inhibiting ROS production by NADPH oxidase or xanthine oxidase. Isoflavan also inhibits the activity of enzymes involved in protein synthesis, such as acetylcholinesterase and phospholipases A2. The experimental model used was the mouse with glabridin induced atherosclerotic lesions due to hypercholesterolemia. In this model, isoflavan reduced lipid peroxidation, inhibited lipid accumulation in macrophages, and decreased vascular smooth muscle cell proliferation.
